Genetic diversity and phylogeny of wild cowpea [Vigna unguiculata L. (Walp)] accessions using Simple Sequence Repeat Markers (SSR)

Cowpea is a highly nutritious grain legume mainly produced in low altitude tropical zones in West African regions. Despite its nutritional importance and high nitrogen content for soil improvement, the production of cowpeas faces many constraints including drought, low yield and diseases and pests’ attacks. The objective of this study was to assess the genetic diversity of wild cowpea accessions to improve cultivated varieties. A total of 34 accessions from diverse geographical locations in Africa were assessed. To characterize the accessions, 61 microsatellite markers were used from which 19 markers were found to be polymorphic. A total of 64 alleles were observed with an average of 3.368 per locus. The analysis of molecular variance (AMOVA) revealed that genetic variation between and within subspecies was 37% and 49%, respectively. The accessions were structured into six populations according to the subspecies with a fixation index (Fst) of 0.199 (p ≤ 0.001) and a strong differentiation between the accessions within the populations. Principal coordinate analysis (PCoA) showed a random distribution of accessions and phylogenetic analysis classified the accessions into three genetic groups. The variability and diversity exist between the wild accessions of cowpea. The wild cowpeas are important in breeding and conservation programs.
